Peptide-Guide: Afamelanotide (M-I)

Posted On: Friday, September 3rd, 2010

http://peptide-guide.com/

Melanotan One, Scenesse, Afamelanotide (M-I) Dose:

Light:  .5mg
Common:  1mg
Large:  2mg

Melanotan/Afamelanotide
is a synthetic version of melanocyte stimulating hormone (a-MSH).  Afamelanotide treatment induces skin pigmentation through melanogenisis, the production of melanin.  Melanin prevents cellular damage in the skin by absorbing, reflecting and refracting light.  Melanotan reduces sun damage to UV exposed skin in those with sun allergies.

Melanotan results from clinical trial

Melanotan Results: Melanotan favors production of eumelanin (black/brown) over pheomelanin (red/yellow) pigment.  Injecting this linear amino acid structure peptide offers super-physiological levels of a-MSH.  Learning the majority of secretion of a-MSH comes from the skin as opposed to the pituitary gland was a recent breakthrough in medical science.  Those with a Fitzpatrick skin type I-III classification have natural levels of a-MSH that are hardly detectable.  The fair skinned can achieve a photoprotective tan with synthetic MSH.

Fitzpatrick skin type chart

Research Use:
The half-life of Melanotan peptides is very brief.  Afamelanotide is being developed in implant form for the ease of use in dermatology.  The implant contains 16mg which has a controlled release effecting melanin density for months.  Melanotan One is offered in research peptide form (20mg Melanotan One).  Peptide Melanotan One gives the user greater control over dosing, however requires injections and expertise.

Lifestyle: Melanocortins have the potential to be the next big thing in lifestyle drugs (Viagra, Botox, etc).  Patients who benefit from treatment have such variations when it comes to their goals, expectations and genetics.  Melanotan can be appropriately used continuously as well as intermittently.  Dose timing and missing doses are generally not of much concern.  Missing a dose and taking time off is the name of the game for the lifestyle peptide.

Recommended Therapy: Intermittent therapy, lasting 10 days (skin type III) to 6 months (skin type I) works well.  Take note how long after your Melanotan administration melanin density stays elevated.

Melanotan and Melanin density

Molecule: Two amino acids present in the bodies natural a-MSH have been changed in production of Afamelanotide creating more stablity.  Melanotan only works when injected subcutaneously.
Amino acid sequence: Ac-Ser-Tyr-Ser-Nle-Glu-His-D-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH2

Approval: Melanotan is still undergoing clinical trials and has not reached approved status.  People use on the pretenses and understanding that it is for research purposes only.  Users may suffer from slight nausea and flushing after subcutaneous injection; however it is reported to be mild and pass quickly.

Safety First: Melanotan One is a selective agonist of the MC1R (Melanocortin 1 receptor) and does not have aphrodisiac effects.  Melanotan One does not cross the blood brain barrier.  It is for this reason Afamelanotide has continued clinical trials.  The fat/weight loss, appetite suppression, libido effects are found in Melanotan 2 and Bremelanotide (PT-141).

Melanotan Before and 3 weeks after comparison photo

Patience: Melanotan One is less efficacious than its counterpart Melanotan 2 for achieving a dark tan.  Afamelanotide will therefore require 8-10x the dosage to see comparable results.  Melanotan One users want minimum side and peripheral effects with a focus on photoprotection.  Be patient and prepared through your Melanotan journey.

Administration of Research Melanotan
Safe and controlled administration of Afamelanotide for research is done much like that of Melanotan 2.  Melanotan One offered as a peptide in a multi-dose sterile vial is desirable.  Implants, nasal sprays and pills are not applicable/efficacious at the present time.

Melanotan Treatment
Allow time before seeing UV exposure.  Sunburn is to be avoided.

Example-  Skin type II, 180lb male
Day 1-2:  .5mg each night
Day 3-6:  1mg when convenient
Day 7-10:  2mg when convenient
Day 11 on forward:  begin dosing ~6mg/week

Day 7:  10min sun bed session.  (Cover face/genitals/sun damage, use lotion, etc)
Day 9:  10min sun bed session.
Day 11:  15min sun bed session

Dosing According to Skin Type

Fitzpatrick Skin Type I:  1mg-2mg/day.  100mg total may be necessary to achieve desired color.  UV exposure after at least 20mg in system
Fitzpatrick Skin Type II:  .75mg-1.5mg/day.  60mg should be sufficient to get color.  UV exposure after 10-20mg
Fitzpatrick Skin Type III:  .5mg-1mg/day.  A sunless tan may be realistic here
Fitzpatrick Skin Type IV and beyond:  Do not use Melanotan

Melanotan Video from Australia

Melanotan can develop a protective tan in users.  A natural tan is developed in response to cell damage caused by UV radiation.  Ultraviolet rays penetrate the upper layers of the skin, triggering a-MSH, stimulating melanin production.  Melanin deposits act as a natural sunblock at the cellular level.  Melanotan tanning injections may offer a way to bring upon a safe, cosmetic tan with less damage.

Reconstituting

Swab the stopper of both your M-I vial and the vial of the dilutent (bacteriostatic water) with an alcohol swab.

Take a syringe and draw up your bact water. The amount isn’t critical, other than making sure you know exactly how much you have used. The best rule of thumb is choose an amount that will make measuring the final product easy.

Example- 2ml(cc) per 20mg vial of M-I would mean each 10 marks on a U100 insulin syringe would equal 1mg of Melanotan (M-I)
Example- 1ml(cc) per 10mg vial of M-I would mean each 10 marks on a U100 insulin syringe would equal 1mg of Melanotan (M-I)

Avoid shooting the dilutent directly on the lyophilized powder.  The powder should almost instantly dissolve.  Gently swirl until the liquid is clear if powder did not dissolve upon contact.

Store your unused M-I in the refrigerator.  Once reconstituted, the Melanotan should be good for 4-6 weeks in the refrigerator.

Measuring

After successfully reconstituting your peptide, now measure the desired amount out for injection. You will want to use a U100 insulin syringe to draw out your M-I.

MT-2: When does the Magic Happen?

MT-2: When does the Magic Happen?

Melanotan 2 has many effects users seek out.  The only synthetic aphrodisiac on the market, the effects of Bremelantide (PT-141) via MT-2 is amazing.  Melanotan 2 can indeed be that magic Barbie peptide which goes beyond Bremelanotide affecting ones facial hair, photoprotection, and weight loss!  A dark sexy beard is often a unique perk for those with light or patchy facial hair.  Appetite reduction, energy and increasing gym productivity are more positives of MT-2.  Then of course, the magic happens when the tan sets in and is dialed in.  Photo-protection through MT-2 is certainly the most impressive characteristic which results in life changing opportunity.

Melanotan 2 and Libido

MT-2 can consciously be used as an advantage in the libido dept.  Good times are right around the corner when used properly.  Be aware that dosing too high too soon leads to a bad time.  Dose escalation and assessing ones tolerance is crucial.

For most users, getting sick and experiencing nausea is not high on the agenda.  To avoid the initial post injection nausea MT-2 users carefully assess tolerance.  Starting extremely low and escalating the dose is wise.  A .25mg dose is very small and for first time users it is advised to dose on a full stomach, an anti-histamine in your system, at around bedtime.  This way a user has the odds on their side that they will experience nothing from the injection  The injection site sometimes itches for a few minutes or can have some discomfort.  A seasoned user will not likely encounter any side effects.

The ultimate synthetic aphrodisiac experience can be seen with proper planned MT-2 use.  When your body can handle and is ready for that 1-2mg dose, 2-4 hours before the encounter…game on!  There are many variables that come into play in regards to PT-141/MT-2/the synthetic aphrodisiac.  Variables such as sex (male/female – works for both), timing, dose, stress/mood and expectations all are factors.

Depending on your objectives, MT-2 can increase one’s libido in a matter of hours or a week or two before realizing the effects.  Many times users really dont pay attention or are aware of the libido properties.  Often men will share that their morning wood is something to take note of.  Women claim the effects are long lasting, up to 3 days after administration.  Others will be kept up at night with desire  Then some can be in sync with their MT-2 libido tolerance and plan a aphrodisiac date night to be remembered ;)

Check out our between the sheets forum specific to the aphrodisiac:

MT-2 Black Beard
For the blond, strawberry, ginger, auburn, etc hair types out there the MT-2 will effect facial hair growing in.  Very rare that hair color will change on your head.  With extensive use it can effect body hair…but again, rare.  Beginning MT-2 users who are loading up, assessing their tolerances and building up their photoprotective skin via MT-2 will start to see the beard coming in darker approximately 2-3 weeks into usage.  Somewhere between 20-30mg a user should be able to see a darker color.

Many enjoy this effect and grow out or sport their shadow with more frequency.  One issue which users should be aware of is that they should still be paying close attention to hygiene & upkeep.  With the adding emphasis on tanning and/or exercise…one shouldn’t necessarily let the facial hair get out of control – even with it looking attractive.  Every now and again users will report an increase in oil.  You want a black beard, not black heads.  Also you want to keep the pores straight and not invite unnecessary ingrown hairs  Flattering compliments about facial hair is not the norm for many Melanotan users, until they experience it!

Rough photo that gives an idea of the darkened facial hair difference:

The Barbie Effect
Melanotan 2 has been coined the barbie drug by the media etc years back as it was found to make one tan, skinny and feel on top of the world (aphrodisiac).  It will indeed decrease your appetite.  Some users use it for this reason as there are more and more studies on melacortins and obesity.  Folks who are not interested in appetite reduction have to work on dosing at appropriate times, be aware of not over consuming when the MT-2 isnt present in the system and dose at the right mg  Some advocate smoking herb before injects to not only hedge any nausea, but to keep the appetite functioning at a high capacity.  Other peptides like HGH and some of the growth hormone anti-aging secretalogues also work to increase appetite and give an edge to their skin/rejuvenation.  Over eating can be factored in according to the desired lifestyle – have yet to see any user logs where anyone gets fat and tan on MT-2.

The combination of the aphrodisiac, the sun light, weight loss, complements…they are all part of the barbie phenomenon.  Users frequently comment about increased productivity in the gym – nothing scientific to back these claims.  Some studies in regards to melancortins and insulin sensivity, etc are out there.  Stay tuned until these are discussed further.

On the flip side users can also find themselves lethargic on MT-2.  If Mt-2 is making you tired, take a few days off from dosing as it will not hurt a thing.  It is all about finding the right Barbie balance

The Magic Tan
As a seasoned Melanotan peptide user, I recall the 5th or 6th week of use being ah ha moment.  There were several epiphanies had during the course of experimentation and experience.  The feeling of having the superior tan (as compared to a spray tan) which was my own was tops.  Beyond that, the experience of being on a lake for extended hours…becoming more tan, not burning…what a thrill.  Freedom to be in the sun and from the sun is a topic I hope to elaborate on in depth.

Most users experience doubt, concern, remorse…all part of the initial roller-coaster.  I know I was crunching numbers, questioning sanity in regards to UV ray exposure, the injections…the buying process…it all sucks.  That 10 day to two week mark where some one calls out your awkward pigmentation is not good for the esteem.  At a month where your tan surpasses the pigmentation…ahhh, good times!  You start to dial in your tan which can resemble that of a quality spray tan.  But now you can continue to live outside, do cardio in the gym and swim without fear of your spray on mess transferring off you and often onto your clothes, bed sheets, and other half.  MT-2 users are able to forget all that and leave it in the past!

Melanotan magic happens through diligence, understanding and appreciation for the skin.  Love and take care of yourself!  Melanotan peptides enhance many lifestyles.  They should not become more trouble than they are worth.  Check out as much info as possible before diving in head first.  Find out each reason you should not use before a decision is made.  Prepare for everything and expect results – these are some of the most efficacious research peptides on the market.

Why Tanning Pills Don’t Work

tanning pills do not work

Why Tanning Pills Don’t Work

Sunless.com

Years ago, the feature Sunless Tanning: Facts and Myths attacked the effectiveness of tanning pills.

But tanning pills have proven to be the hardest sunless tanning myth to kill. Nutritional supplements, or tanning pills, that “promote” tanning proliferate, and ads for them are everywhere.

There are only two things that matter in the production of melanin: your genetics and your degree of exposure to ultraviolet radiation. Nutrition simply does not play a part for the normal, well nourished individual.

To make this clear, I’ll go over the facts of tanning [melanocytes - melanogenesis], describe three medical conditions that negatively effect tanning, summarize drugs that can affect melanogenesis, and, finally, analyze three typical tanning pills.

Melanotan II Instructions

A compilation of information regarding how to use MT-II for research. MT-II is an analog of the peptide hormone alpha-melanocyte stimulating hormone (α-MSH). This hormone acts on melanocytes to stimulate melanin production. Melanin is your pigment responsible for your tan. MT-2 out competes a-MSH for binding to melanocyte receptors in humans and other mammals because it has a binding affinity over 1000x. This is the reason why the fair skinned CAN reach a natural tan.

Please pursue information regarding the usage of these products from your own research, academic journals, or the research of your in-house scientific team. Products are sold for research purposes are not for human consumption. Remember when you contact these types of stores/sponsors not to relate the questions or topics to human consumption or they tend not to answer. Always inquire in a research type question. Cause after all, we know that this is for your experiments and not for human consumption.

Mixing: Add bacteriostatic water to the vial when you are ready to begin journey. The peptide starts to degrade after being reconstituted.
Comparable homemade video:

I recommend adding 100 units of water to the vial. This will minimize the volume that you have to inject and will simplify the arithmetic.

1ml syringe, 1ml bact water to reconstitute
Calculations (given you want a .5mg dose):
step 1= 1ml
step 2= 10mg MT-II
step 3= 1ml bact water
step 4= 500mcg dose
2-3 ticks on your insulin pin (1/20th a syringe full)

peptide calculator image
Some people prefer to add more dilutent which works fine, take note of the volume increase.

Needles: 29-31 gauge X 1/2″, 1 CC (100 unit). That is a typical insulin needle. Once you get your technique perfected, it is almost painless. More info

Starting dose: Your first injection should be a very small dose, for example .25mg. See how you react. Goal should be to feel nothing. Dose after dinner, before bed. Any dosing chart stating that you should take a high dose (according to your weight) is outdated and potentially dangerous.

Loading dose: I recommend loading with 0.5-1mg once a day. People who have used doses in this range generally report getting excellent results. Don’t worry if you miss occasional days. It will not make much difference, focus on the cumulative effects. Nausea and yawning were frequently reported side effects due to Melanotan II

Maintenance dose: Maintenance is taking doses less frequently than daily to avoid becoming darker than you want. Yes, that will happen. With enough UV, you will get much darker than you have even been before. I recommend .5mg every mon. and thurs. In my case, I tan on Weds. in between the two doses, then in the weekend during the summer I am in the sun. It works great.

UV Radiation: MT-II is not a good sunless tanner. You need UV (from sun or a tanning bed) to develop a tan. Without UV almost nothing happens. In other words, NO UV = NO TAN. Well, user will pigment…. But if you have loaded for a full month and then start UV exposure, you (and your friends) will be astounded by how fast you tan and how dark you get.

Watch yourself: Your tan can sneak up on you. A tan generally sets in 3 days after UV rays

Avoid burning: You are protected from burning mostly by your tan, not the melanotan II that you have taken. Therefore, don’t overdo the rays at first. Start with only as much UV that you could tolerate without burring before you started melanotan II. Then gradually increase your exposure. It shouldn’t take many weeks before you can tolerate many hours of strong sun without burning…which truly is incredible for those who have never experienced!

Continue your regular daily dose until you have reached your desired tan and do not want to become darker. Then cut injection frequency to once every 2, 3, 4, or even 7 days. You will have to experiment to find the frequency that gives you the tan that you want.

After mixing with water, it should be refrigerated. Not a problem to store 8 weeks

Melanotan:
melanotan freeze dried peptide photo

Storage: It is a very durable peptide. Before it is mixed with water, melanotan II is quite stable and can last for months at room temperature.

Do you have to inject MT-II?
Yes. The best, most efficient method of administering melanotan is via sub-q injection. Nasal sprays do not work.
tanning injection

Note: There are many things you will need to consider before experimenting with this peptide. MT-II is not an approved drug (anywhere to my knowledge). MT-II is legal to buy, possess, etc. Variables such as skin type and individual goals need assessment, create a user log. When you start using melanotan to tan keep in mind that tanning is literally a side effect, not the objective. Melanotan is not going to color your skin, it is going to make your own skin create its own tan and that in turn creates UV protection. If you are looking to be some bronzed beach god with perfectly uniform and specific color then you are better off to going to mystic tan.

Know your skin type: Knowing your skin type will help you create a user log. There are 10s of thousands of melanotan users worldwide who share the experience :)
fitzpatrick skin type chart
Melanotan is best suited for the folks with skin types I & II

How should I dose MT-II?
Start out with small doses and build up. A typical starting dose is around .25mg and max dose reaching 1mg. You can look at it as being as simple as that.

Things to consider:
There is no magic pill or formula. Few dermatologists know about melanotan. The skin is a large, unpredictable organ. Feel comfortable and confident with MT-II before use. Check out as many before and after photos and user logs as you can.
melanotan before and after photograph

How much should I buy and how long will it last?

Skin type I: 30-50mg
Skin type II: 20-30mg
Skin type III: 10mg
Should last entire summer or season

Melanotan and finances?

Is melanotan cost prohibitive? Absolutely. There is no reason any melanotan user should be on a budget. At the same time, MT-II is a valuable investment for the prime candidate.
Initial investment can be $150-200 for product, $90 for tanning salon (you know the fees, commitment, lotions, etc add up). HOWEVER, melanotan has a lasting effect.
As a user for the past 4 years I will paint a rough picture of my expenditures as I remember them:
2005: 30mg first purchase, tanned in the sun…learned a bit about my skin type I body
arrived at a tan 3-4 weeks later and was blown away. Must have bought another 20mg and spent $300 total
2006-2007: Probably invested in 100mg over the course of those two years…vacationed a lot and spend ~$600 on melanotan and brief months at the salon
2008: 30-40mg used, very small doses required for results this year ($200?)
2009: 20-30mg …so maybe $100
Future I anticipate using 1-2 vials/yr.

How soon will I begin to see results?

You should notice a change in your skin tone after three to four weeks. If you have freckles, expect them to get darker before your actual skin color changes.

How long will tan last?
A tan developed using melanotan II lasts much longer than an ordinary tan. A well-tanned person returning from a beach holiday will lose most of the tan in a month if they stop getting sun. But if they had been using melanotan II and continued on maintenance after returning, they would still have most of their tan 3 months later.

Side effects of MT2?

Possible short-term side effects you should know about, including: nausea, appetite loss, facial flushing and increased libido. These may be noticeable during the first few days of treatment but should taper off.

My Guide to MT-II: Avoiding sides with best possible results
tan bodybuilder
By: Jay

First off, let me start by saying MT-II has been nothing short of a miracle for me. I have NEVER had a good tan like i have had since using it…in my ENTIRE life.

Sides

Most commons sides that pose a problem tend to be: nausea, loss of appetite, headache, and indigestion

Other common not so problematic sides are: frequent erections, flushing on first couple doses, darkening of freckles and moles (does go away after use in most cases).

Dosing: Loading

Loading is important BUT do not overstep your limits. My first ever MT-II cycle i hopped right on at 1mg ed and had no problems. However, the last go around i did that and was sick with no appetite for a week. The key is, take it slow. I would work it like this:
